Professor Li Chuanyou's team from Shandong Agricultural University has made important achievements in the field of plant regeneration. The team found that REF1, a regenerative factor produced by cell injury, is an original injury signaling molecule that triggers tissue repair and organ regeneration. This discovery will bring significant application value to the field of plant gene research.

Cell damage is the primary physical inducement that triggers plants and animals to start the regeneration process. In the 1970s, scientists discovered that micropeptide signaling system and the plant hormone jasmonic acid regulate systemic defense responses in plants through a common signaling pathway.

In subsequent studies, the scientific community has a deep understanding of the signal transduction mechanism of plant systemic defense, but the mechanism of plant damage repair and organ regeneration is not well understood.

Professor Li Chuanyou's team has been studying tomato for a long time, using genetic means to analyze the plant systemic defense signaling pathway jointly regulated by systemins and jasmonic acid. After years of research, the team identified one of a series of tomato mutants with changes in systemin signaling pathways that showed deficits in both defense and regeneration.

In follow-up studies and comparative experiments on this mutant, the researchers identified the key role of a small peptide that was applied externally to significantly improve the tomato's regenerative capacity.

The team named it regeneration Factor REF1. Regeneration factor REF1 regulates the process of plant regeneration, including the recognition of receptor proteins, transcriptional activation of cell reprogramming regulatory factors, and further amplification of REF1 signals, etc., which have been confirmed by studies.

Using the regeneration factor REF1, the research team also found that in the plant kingdom, REF1 small peptides and their receptors are widely distributed in dicotyledon and monocotyledon plants. REF1 application can not only significantly improve the regeneration ability and genetic transformation efficiency of tomato,

but also increase the regeneration ability and genetic transformation efficiency of soybean, wheat and corn several times. This provides a convenient and universal solution to solve the bottleneck problem of low efficiency of crop genetic transformation in biological breeding.
